LncRNA and Infection Biology

RNA is a truly remarkable molecule with functions and activities far beyond that of an intermediate information carrier. The abundant class of long non-coding RNAs (lncRNAs) contains highly specialized RNA with structural or regulatory functions that range from assembling large protein complexes to localizing, sequestering, or allosterically modifying proteins and other interaction partners. Our genome contains thousands of lncRNAs, many of which are specifically regulated during bacterial or viral infections. However, their contribution to launching and sustaining an effective host response remains elusive. Our group combines a cutting-edge suite of technologies from the fields of biochemistry, genomics, molecular biology, and computational biology to decode how lncRNA work mechanistically and how they contribute to host defense mechanisms. This group is located at the Helmholtz Institute for RNA-based Infection Research (HIRI).

Molecular Principles of RNA Phages

The research group led by Jens Hör investigates the dynamics of infection, host takeover, and anti-phage defense during the interaction between RNA phages and their hosts. They seek to understand the molecular principles underlying these processes to develop new and improved antibacterial strategies such as phage therapy.

Host-Pathogen-Microbiota Interactions

The importance of RNA in maintaining cellular physiology by controlling gene expression in response to intrinsic and external cues has long been underestimated. Now, numerous human diseases have been linked to RNA functioning. Likewise, we now know that bacterial pathogens harness a large suite of noncoding RNA molecules to adapt to environmental stress and to precisely regulate their virulence programs. In an era of antibiotic crisis, it is essential to discover alternative combat strategies against pathogenic bacteria — ideally ones that spare the beneficial microbial species. The high specificity of RNA molecules provides great potential for achieving these goals. This group is located at the Helmholtz Institute for RNA-based Infection Research (HIRI).

Genome Analytics

The Genome Analytics (GMAK) facility provides next-generation sequencing (NGS) technology to the HZI. External users may also have access to the technology, e.g. upon collaborative arrangements. In addition to sample DNA/RNA quality check, library preparation and sequencing, the facility provides primary data processing, data quality check and analysis. A number of pipelines for secondary data processing are established.

One Health Surveillance

The One Health Surveillance Core Unit facilitates the comprehensive long-term collection of samples and data on human, animal and environmental health. This sample collection serves as the basis for research in HIOH‘s departments, as well as for collaboration with our partners.

Natural products from underexplored pathways and extreme environments

Drugs with new chemical entities are urgently needed to tackle the emerging antibiotic resistance. We investigate natural products from underexplored pathways and extreme environments to characterize bioactive molecules with true structural novelty and their biosynthetic enzymes to bioengineer and improve potent anti-infectives.

Central Facility for Microscopy

Light microscopy and electron microscopy are excellent high resolution tools to investigate the adhesion and invasion mechanisms of pathogenic bacteria. High resolution electron microscopy permits to examine and unravel pathogenicity mechanisms on a morphological basis. ZEIM offers equipment and highly sophisticated expertise for the preparation and observation of biological samples.

Epidemiology

Epidemiology conducts research on health and disease at the population level – infection epidemiology is concerned with contagious diseases. Their tools and methods are systematic queries, clinical examinations and laboratory diagnostic documentation for both healthy and afflicted individuals, as well as statistical analysis of the compiled data. Causes and risk factors for infections can thus be identified. Infectious diseases epidemiology contributes to the development of preventive measures, early detection and therapy for diseases. Moreover, it examines the efficacy of such measures. Thus epidemiology ties in with scientific findings in basic research as well as medicine, and examines these processes at the population level.

Experimental Infection Research

If we are attacked by a virus, the immune system reacts within a matter of hours. Highly specialized immune cells recognise the pathogen and release highly efficient messengers, which activate the immune system. These messengers include the interferons, which ensure that individual host cells are mildly infected . At the same time, interferons can also influence the course of the immune response and the memory of the immune system. Without these messengers virus infections - which we normally overcome almost unnoticeably - become fatal within just a few days. The group Experimental Infection Research is based at the TWINCORE in Hannover.

Core Facility of Comparative Medicine

To be able to investigate diseases, understand the relationships between pathogens and their host and to develop novel strategies against pathogens, the scientists at the HZI depend on experiments that involve the use of laboratory animals. For this purpose, the HZI operates a central animal facility on campus. The primary task of this service unit is the breeding and keeping of laboratory mice as well as provision of services for the scientists of the HZI, centered around laboratory animal science.
